Purpose: Filter 250 mls of new media and add 10% FBS (practice filtering media)

Materials: 

250 mls Fresh Media made from Powder

25 mls Fetal Bovine Serum

2.5 mls anti-biotic/anti-mycotic (Penn/Strep)

Nalgene Bottle Filter

5 ml Pipette

25 ml Pipette

Pipette Aid

Vacuum Pump

70% EtOH

Procedure: 



• Sprayed everything down with 70% EtOH before entering the hood. Sprayed hood surfaces down too. Hood had been running fan and UV light for approximately 15 minutes before use. 
• Unwrapped Nagene Bottle Filter and added 250 mls of media, 25 mls of FBS, and  2.5 mls of anti/anti. 
• Attached the hose to the filter and pumped media through filter.
• Removed top and filter portion of bottle filter, then capped and labeled the new media bottle.
• Cleaned up hood, put used pipettes in biohazard and put supplies away.
