Solution Preparation: Tris

Purpose: Prepare solutions to use throughout the rest of the term for Protein Purification class.  Making 1M Tris pH 7.6.



Materials: 

1L 1M Tris: 121.14g Tris (FW: 121.14g)

diH2O (4 degrees C)

1L Glass Bottle

Lab Tape

Balance: Ohaus Voyager V16120

Weigh boat

1L Graduated cylinder

pH Meter: Accumet AB15

6M HCl



Tris: Fisher Tris Base (FW 121.14g), Lot 091721



Procedure: 



1. Weighed out 121.14 g Tris Base in weigh boat

2. Set up 500mL Beaker on stir plate with stir bar

3. Added approximately 250mL cold diH2O to 500mL beaker

4. Added 121.14g Tris base

5. Turned on stir plate to stir Tris into solution

6. Calibrated pH Meter

7. Took initial pH reading of solution

8. Added 6M HCl to pH to 7.6

9. Transferred solution to 1L graduated cylinder

10. Rinsed beaker with diH2O 

11. Used rinse water in beaker to add to volume in graduated cylinder

12. Brought to Volume in graduated cylinder at 1L
