Solution Preparation: Z Buffer
Purpose: Prepare solutions to use throughout the rest of the term for Protein Purification class. Making Z Buffer (without DTT).
Materials:
For 500mL:
4.26 g Na2HPO4 (FW 141.96)
3.17 g NaH2PO4*H2O (FW 137.99)
0.373 g KCl
0.123 g MgSO4*7H2O (FW 0.373)
5mM DTT (Added later based on how much ZBuffer aliquot being used.)
Stir bar/Stir plate
1L Glass Bottle
Lab Tape
Balance: Ohaus Voyager V16120
Weigh boat
1L Graduated cylinder
Procedure:
1. Weighed out:
4.26 g Na2HPO4,
3.17 g NaH2PO4,
0.373 g KCl,
0.123 g MgSO4*7H2O
2. Poured approximately 400mls of diH2O in beaker with stir bar.
3. Started stir plate on low setting.
4. Slowly added chemicals in the order listed as weighed out.
5. When entirely in solution, transferred to graduated cylinder and BTV @ 500mL.
6. Labeled as: "Z Buffer w/Out DTT 02/13/13 JNSA"
